SPRINTseq In-situ Sequencing

Signal readout for SPRINTseq. Perform after RCA (and optional autofluorescence quenching).

Flow Cell Assembly

• Assemble slide into Flow Cell at this stage (if not done earlier).
• Ensure all tubing connections are leak-free.

Sequencing Cycles

• In-situ Sequencing: Complete all sequencing cycles (typically 10 cycles) using two-color reversible terminator SBS chemistry.
• Detailed operational instructions are available in the internal documentation (Docker build).

Detailed public sequencing protocol is being prepared. Contact the lab for current reagent specifications and imaging parameters.
