Huang Lab · Peking University
Spatial transcriptomics, from bench to analysis.¶
Yanyi Huang Group · BIOPIC & ICG
Bench-ready protocols, complete analysis code, and reproducible workflows for PRISM and SPRINTseq — two padlock probe-based spatial transcriptomics technologies.
Technologies¶
Two methods, one shared workflow -- diverging only at readout.
-
PRISM
High-plex spatial RNA imaging in a single round using color-intensity barcodes and radius vector encoding. Up to 64-plex with conventional microscopes.
-
SPRINTseq
Rapid, signal crowdedness-robust in-situ sequencing through hybrid block coding. Profiles a targeted transcriptome in under 9.5 hours.
What You Will Find¶
-
Bench-Ready Protocols
Not just "methods" sections -- step-by-step SOPs with exact reagent volumes, temperatures, and timing.
-
Bills of Materials
Complete reagent lists with catalog numbers, vendors, and preparation guides.
-
Analysis Pipelines
From raw images to expression matrices -- registration, spot detection, decoding, and segmentation.
-
Troubleshooting
Real-world solutions to common pitfalls from years of lab experience.
